I lose it when Kirk silently mouths "his brain?!?" while squinting to indicate his utter noncomprehension. Just another masterstroke from a consummate actor.

As funny as "He's dead, Jim" always was, my favorite Trek quote has to be from SNL, when Shatner was on back in '86... Phil Hartman as McCoy:
Kirk: "Bones, this man needs medical attention!"
McCoy: "Dammit, Jim, I'm a doctor, not a -- oh, right."
